I h...I've been meaning to comment for a while. I have done four multi-day charity rides...300+ miles with days of 75-100 miles. I don't remember what kind of bike you have, but what made a huge difference for me was getting a bike fitting...about $100 and a new saddle. It totally goes against logic, but the smaller and less padded the saddle the more comfortable you'll be. Another tip...butt balm. Google it.
